A.  Personal Statement:
    The long-term goal of my laboratory is to help develop clinically-useful drug therapies that will ameliorate or even restore cognitive and autonomic functions in patients with age-, disease- and trauma-related impairments. The prime targets of our research are the central nervous system, central neurons and ligand-gated ion channels, such as nicotinic acetylcholine receptors (nAChRs). My laboratory uses integrative, multidisciplinary approaches and state-of-the-art experimental tools including patchclamp electrophysiology in brain slices and acutely dissociated neurons, fluorescent Ca2+ imaging, in vivo and ex vivo fluorescent tracing, behavioral models (e.g., Morris water bath, brain trauma), RT-PCR, Western Blot and immunohistochemistry to delineate molecular and cellular mechanisms and network properties contributing to performance of functionallydefined brain circuits. The main focus is placed on a7 nAChRs and how these sources of Ca2+ entry support Ca2+ homeostasis, neuronal function and survival. Collectively, these research efforts may help develop novel therapeutic strategies for selective targeting of specific pathways supporting cognitive and autonomic functions in health and disease.
